Press Ctrl+Alt+Shift+S to save the roll over button. Save as an HTML file along with images. Photoshop will take care of the java script code that is required for rollover button to function.

In Dreamweaver MX, on the Common bar, click the Image icon, and choose "Rollover Image". Put the two images you've made as the Original Image and the Rollover Images.

4. Now turn of the visibility by clicking on the eye to the left of the button_down layer. Go to File > Save for Web and save the image.
